          摘要:目的 建立HPLC同時(shí)測(cè)定紅花藥材中羥基紅花黃色素A、蘆丁、槲皮素、山柰素4種化學(xué)成分含量的方法。方法 采用Agilent ZORBAX SB-C18色譜柱(4.6 mm×250 mm,5 ?m),DAD檢測(cè)器,以0.5%磷酸溶液(A)-甲醇(B)進(jìn)行梯度洗脫,柱溫為30 ℃,流速為0.8 mL/min,進(jìn)樣量為10 ?L,檢測(cè)波長(zhǎng)為360 nm。結(jié)果 羥基紅花黃色素A在0.077 6~0.776 0 ?g(r=0.999 9)、蘆丁在0.046 0~0.460 0 ?g(r=0.999 6)、槲皮素在0.006 4~0.064 0 ?g(r=0.999 9)、山柰素在0.012 8~0.128 0 ?g(r=0.999 7)范圍內(nèi)呈良好線性關(guān)系。羥基紅花黃色素A、蘆丁、槲皮素、山柰素的平均回收率分別為99.68%、99.78%、102.03%、97.03%,RSD(n=6)分別為2.29%、1.52%、1.02%、2.05%。結(jié)論 該法簡(jiǎn)便、準(zhǔn)確,靈敏度高,穩(wěn)定性和重復(fù)性好,可用于紅花藥材的質(zhì)量控制。

          Simultaneous Determination of Four Chemical Components in Carthamus tinctorius L. by HPLC HE Ting1, LI Ke-ao2, JI Zhi-hong2, TIAN Shu-ge3 (1.           Abstract: Objective To establish an HPLC method for the simultaneous determination of hydroxysafflor yellow A, rutin, quercetin, and kaempferol in Carthamus tinctorius L.. Methods The Agilent ZORBAX SB-C18 column (4.6 mm × 250 mm, 5 ?m) was used with diode array detection. The mobile phase was 0.5% phosphoric acid (A) and methanol (B) to separate the four components by gradient elution at 30 ℃; the flow rate was 0.8 mL/min; the injection volumn was 10 ?L; the detection wavelength was at 360 nm. Results The linear ranges of hydroxysafflor yellow A, rutin, quercetin, and kaempferol were 0.077 6–0.776 0 ?g (r=0.999 9), 0.046 0–0.460 0 ?g (r=0.999 6), 0.006 4–0.064 0 ?g (r=0.999 9) and 0.012 8–0.128 0 ?g (r=0.999 7), respectively. The average recoveries of four components were 99.68% with RSD=2.29% (n=6), 99.78% with RSD=1.52% (n=6), 102.03% with RSD=1.02% (n=6), 97.03% with RSD=2.05% (n=6), respectively. Conclusion The method is convenient, accurate, sensitive, stable and repeatable, which can be a method for quality control of Carthamus tinctorius L..

          紅花為菊科紅花屬植物紅花Carthamus tinctorius L.的干燥花,始載于《開寶本草》,在我國(guó)已有多年栽培和藥用歷史,主要分布在新疆、河南、四川、云南、浙江等地[1]。紅花具有活血化瘀、散瘀止痛、降血壓和降血脂等功效[2]!侗静菥V目》記載,紅花能“活血潤(rùn)燥,止痛散腫,通經(jīng)”。臨床和藥理研究發(fā)現(xiàn),紅花對(duì)心腦血管系統(tǒng)、神經(jīng)系統(tǒng)、免疫系統(tǒng)等均可發(fā)揮一定作用,同時(shí)具有抗炎、鎮(zhèn)痛、抗腫瘤、抗菌、抗氧化等多種生理活性[3-5]。
          紅花的化學(xué)成分有黃酮、生物堿、聚炔、有機(jī)酸和多糖等,其中以黃酮類化合物(紅花黃色素)為主要有效成分[6-9]。2015年版《中華人民共和國(guó)藥典》[10]僅對(duì)紅花中的羥基紅花黃色素A和山柰素進(jìn)行了含量測(cè)定,未對(duì)其他成分進(jìn)行研究。為更好開發(fā)和合理利用紅花資源,本試驗(yàn)以新疆紅花為研究對(duì)象,采用HPLC同時(shí)測(cè)定紅花藥材中4種化學(xué)成分含量,為有效控制藥材質(zhì)量、提高紅花質(zhì)量標(biāo)準(zhǔn)提供依據(jù)。
